Am. Chem. Soc. 12 篇,Angew. Chem. Int. Ed. 26 篇。其中金属室固定成员发表了 122 篇论文,包括 J. Am. Chem. Soc. 5 篇,Angew. Chem. Int. Ed. 12 篇,Chem. Soc. Rev. 3 篇 , Acc. Chem. Res. 1 篇 , Chem. Sci. 1 篇 , Adv. Synth. Catal. 10 篇 , Chem. Commun. 10 篇,Chem. Eur. J. 5 篇,Org. Lett. 18 篇,J. Org. Chem. 3 篇, Organometallics 7 篇等。 实验室今年在金属有机配合物合成、结构及反应化学、不对称催化、过渡金 属催化的新反应等方面取得了一些突出的成果: 1. 在金属有机配合物合成、结构及反应化学的研究方面,成功地合成了第 一例稀土金属末端氮卡宾化合物-钪末端氮卡宾化合物,研究了其反应性能和反 应模式 (Chem. Commun. 2012, 48, 3403);以 2-三丁基锡基呋喃作为金属转移试 剂,实现了金催化的 1,6-二炔-4-烯-3-醇环化/金属转移反应,并通过呋喃锡与阳 离子型金络合物的当量实验分离得到的二金物种直接证明了金属转移反应的发 生,同时也揭示了金催化的反应体系中可能广泛存在二金中间体(Angew. Chem. Int. Ed. 2012, 51, 6181);深入研究了 In-TOX/Ni(II)催化下的不对称合成反应,通 过三噁唑啉配体 In-TOX’与高氯酸镍的配合物单晶,研究了边臂上单噁唑啉在催 化体系中的重要作用,发现边臂上的手性基团对反应的活性以及对映选择性控制 有着重要的影响 (J. Am. Chem. Soc. 2012, 134, 9066)。 2 2. 在不对称催化的研究方面,通过 SpinPhox/Iridium(I)络合物催化 α, α’-二 (2-羟基亚芳基)酮的不对称催化氢化-缩酮化,首次实现了芳香螺缩酮化合物的催 化不对称合成,该反应在温和的条件下,能够以高的收率、优秀的非对映和对映 选择性得到目标化合物,工作发表在 Angew. Chem. Int. Ed. 2012, 51, 936 上,并 先后被 SYNFACTS 和 Angew. Chem. Int. Ed.积极评价;进一步研究了过渡金属铱 催化的去芳构化反应,将铱催化的不对称烯丙基去芳构化反应成功地运用到了取 代吡咯中(Chem. Sci. 2012, 3, 205),并通过铱催化的吲哚体系分子内烯丙基去芳 构化实现了具有 5 元螺环结构骨架的 Spiroindolenine 衍生物的不对称合成 (Angew. Chem. Int. Ed. 2012, 51, 1680)。 3. 在过渡金属催化的新反应方面,研究了两价钯催化的通过芳烃 C-H 活化 启动的串联环化反应或碳-碳偶联反应;研究了 CuBr 催化和 ZnI2 促进的轴手性 联烯的不对称合成,筛选了一系列仲胺,发现当使用吡咯烷时,通过简单的过滤 操作后再加入 ZnI2 即可成功实现从炔丙基胺到联烯的转化;深入研究了金催化 下的羟甲基亚甲基环丙烷的分子内环化反应和亚乙烯基环丙烷的开环反应,并对 张力分子在金催化下的 C-C 键的断裂模式 (Chem. Soc. Rev. 2012, 41, 3318) 和亚 甲基环丙烷张力分子的反应新模式(Acc. Chem. Res. 2012, 45, 641)进行了总结;研 究了钯催化的活性烯烃氧化芳三氟甲基化反应,以活性烯烃为底物,TMSCF3 为三氟甲基源,在室温条件下以较高收率得到芳三氟甲基化产物,从而构建含三 氟甲基的氧化吲哚骨架并进行了初步的机理研究(J. Am. Chem. Soc., 2012, 134, 878)。 今年实验室的重大研究成果有: 1. “基于边臂策略的立体化学控制与催化反应研究”项目获国家自然科学二等 奖 现代合成反应要求经济、高效、环境友好的化学。发展新型催化剂(试剂)以 实现选择性控制与提高催化效率成为现代有机合成化学研究中最重要和最具挑 战性的领域之一。“基于边臂策略的立体化学控制与催化反应研究”围绕叶立德反 应和不对称催化中选择性控制与催化问题,在科技部“973”、国家基金委重点项 目、杰出青年基金和创新研究群体、中科院“百人计划”等项目资助下开展了系统 的创新性研究。项目运用“边臂策略”设计新的催化剂(试剂),成功应用于高选择 3 性的有机合成反应和功能分子的合成中,发展了一些调控有机合成反应选择性和 提高催化效率的新策略、新方法及新反应,为新型催化剂(试剂)的设计和反应 选择性控制提供了新思路和途径: (1) 采用“边臂策略”首次设计合成了假C3-对称性手性噁唑啉配体(TOX), 发现TOX/ 金属体系在傅-克反应、Kinugasa 反应、环加成与环丙烷化反应等多 个重要的不对称催化反应中表现出对映选择性提高、配体加速、杂质容忍性好和 催化剂用量低等优异性能,阐明了“边臂”的作用;首次实现了Cu(II) 催化的不 对称Kinugasa 反应;首次通过反应条件的改变实现了吲哚和硝酮对亚烷基丙二 酸酯的不对称傅-克反应和环加 成反应立体选择性的调控。该策 略和配体多次被国际同行应用于 不对称催化反应中。 (2) 高效催化和高选择性控制 是叶立德化学面临的挑战性课 题 。 利 用“ 边 臂 策 略 ” 设 计 了 PEG-Te 催化剂,将叶立德烯基 化反应的催化剂用量降至最低0.5 mol %,是迄今为止叶立德反应催化剂效率最 高的例子 ; 利用“羟基”或“锂离子”边臂经叶立德途径实现了乙烯基环丙烷化合物的 多样性和高选择性的直接合成,发展了调控该类反应立体化学的新方法和新策略, 并实现了催化的不对称叶立德环 丙烷化反应;发现了一些叶立德 启动的新的串联环化反应,为合 成杂、并环化合物提供了新途径。 这些方法被成功地应用于一些生 物活性分子的全(表观)合成中。 (3) 利用"边臂策略"设计 的新型吡咯烷-脲双功能催化剂 实现了高效催化的环酮对硝基烯的Michael 加成反应(最高达dr > 99/1 和98% ee)。该催化剂被国际同行成功应用于(+)-trachyspic acid 的全合成中。 4 研究成果在多种国际重要化学期刊发表,包括Acc. Chem. Res.1 篇, J. Am. Chem. Soc. 7 篇,Angew. Chem. Int. Ed. 2 篇,影响因子> 9.0 的12 篇,所发展 的催化剂和手性配体被国际同行成功应用于有机合成反应和功能分子的合成。20 篇核心论文的总影响因子202.9 (以2010 年计),篇均影响因子10.1;SCI 他引 1070 次,篇均他引53.5 次,单篇最高他引183 次。培养了15 名博士,其中1 人获中国科学院院长特别奖和全国百篇优秀论文,1 人获中国科学院50 篇优秀 论文和全国百篇优秀论文提名奖。这15 名博士中,目前1 人入选中国科学院百 人计划,5 名已成为985 高校教授或副教授(其中1 名入选青年千人)。项目负 责人应邀在包括IUPAC 国际金属有机等系列会议上作邀请或大会报告11 次,在 双边会议上做邀请报告5 次,2002 年获国家杰出青年基金,2004 年获Thieme Journal Award,2006 年获第九届中国青年科技奖。本研究丰富和发展了有机合 成化学中选择性调控和催化的方法和途径,在一定程度上推动了有机合成化学的 发展,项目主要成果处于国际领先水平。 2. “手性膦氮配体的设计合成及在不对称催化中的应用” 项目获上海市自然科 学一等奖 手性化合物在生命和材料等领域的地位举足轻重,不对称催化则为获得手性 物质的最理想方法之一。金属催化不对称反应中配体的作用至关重要。 该项目 从手性配体作用的认识和不对称催化过程的理解着手,设计合成和改造了一系列 手性膦氮配体,围绕不对称催化一些反应中的挑战,重要不对称催化新反应的实 现以及反应选择性调控、配体中多手 性因素的相互关系等开展研究,取得 了如下重要成果: ( 1 )设 计 了 新 型 膦 氮 配 体 SIOCPhox,对类型广泛的单取代烯丙 基底物实现了高区域、 高对映选择性, 为国际最好结果;实现了开链酮、酰 胺等羰基 α-碳负离子等“硬”碳负离子 的高对映选择性烯丙基烷基化反应,成果具国际领先;实现了钯催化高选择性不 对称环丙烷化反应,为该领域一个重大突破;实现了对碳亲核试剂的动力学拆分, 5 提供了简便获得各种光学活性化合物的新途径。 (2)首次实现了铜催化硝基烯 烃与甲亚胺叶立德的不对称 1,3-偶 极环加成反应和铱催化 α,β- 不饱和 酮碳碳双键不对称氢化。实现了铜 催 化 不 对 称 1,3- 偶 极 环 加 成 及 Mannich 反应的非对映选择性调控 和钯催化不对称 Heck 反应中产物构 型调控,解决了 Mannich 反应中难 以实现 anti-选择性的难题。 (3)阐明了二茂铁配体中 平面手性对于不对称诱导的作 用及其作用方式。 相关成果已在重要国际刊 物上发表。20 篇重要论文被他 引 1154 次,单篇最高他引 210 次。编著专著一部。 5 项发明专 利获授权。在重要国际会议做 大会报告、邀请报告 10 余次。 所获成果对有机化学特别是不对称催化领域的发展起到了重要推动作用。 二、 队伍建设与人才培养 实验室的学术骨干队伍以中国科学院院士、杰出青年基金获得者和中科院百 人计划入选者为主体。现有在职人员 81 人,副高级以上研究人员 38 人,这其中 包括研究员 15 名(含院士 3 名),国家杰出青年基金获得者 8 名,科学院“百人计 划”入选者 8 名,“青年千人计划”入选者 1 名,“千人计划(创新人才短期项目)” 入选者 1 名。 2012 年实验室固定人员中余金权研究员获 “千人计划(创新人才短期项 目)”;刘国生研究员获 “国家杰出青年基金”;邓亮研究员获 “优秀青年科学基 金”;丁奎岭研究员获 “上海市优秀院所长奖上海市优秀院所长奖”; 唐勇研究 6 员获“ 黄耀曾金属有机化学奖” ;游书力研究员获“Asian Core Program Lectureship Award (Japan, Singapore)” 、“AstraZeneca Excellence in Chemistry Award ”、 “中 国科学院优秀导师奖”、 “中国科学院研究生院 BHP Billiton 奖学金导师科研奖”。 2012 年实验室在读硕士研究生 34 名,博士研究生 72 名,在站博士后 3 名; 毕业硕士 4 名,博士 23 名。 三、对外开放与学术交流 今年实验室面向所外单位设立的开放课题有 4 项,资助总额 130.6 万元,均 按照预定的研究目标顺利执行,由开放基金资助完成的优秀成果如下: 优秀成果(一): 首次报道了以炔丙基叠氮和酸为底物,通过直接的氧化脱氢偶联简洁高效的 形成炔丙基三级碳的碳氧键的方法,该方法条件温和,避免了卤代物的使用,具 有很高的原子经济性,且通过控制实验证明,叠氮在这一偶联中可能是一种重要 辅基。得到的含有炔,叠氮和酯的多官能团产物是一种重要的合成中间体,它可 以进一步转化成三氮唑, 苯并三氮唑和烯基醛等可能具有生物活性小分子(Angew. Chem. Int. Ed. 2012, 51, 10823)。 优秀成果(二): 在 C-O 活化的研究方面,首次实现了苄醇 sp3 C-O 的在 Fe,Co,Ni 等催化 下与格式试剂的偶联反应,并通过机理研究进一步实现了从醇到格式试剂的转化 (J. Am. Chem. Soc. 2012, 134, 14638)。 优秀成果(三): 利用双锂试剂的“协同效应”,建立了高效合成氮杂半瞬烯分子的2种方法, 首次测定了该类分子的活化能和单晶结构,发现了该类分子的多种化学反应类型, 该项研究结果表明该类分子存在中性同芳香性,为进一步的实验研究奠定了基础, 该工作被选为JACS Spotlights。 学术交流对于提高重点实验室的研究水平具有重要意义。实验室今年分别接 待了美国、日本、德国、英国、加拿大、西班牙、瑞典等国以及包括中国香港、 台湾在内的的 41 位科学家来室访问进行了学术交流。同时,实验室学术带头人 积极参加国际国内的重要学术会议,应邀在国内外学术会议上作邀请及大会报告 7 39 人次。 2012 年 10 月 9-12 日,由金属有机化学国家重点实验室主办的第五届国际 均相催化论坛(The Fifth International Forum on Homogeneous Catalysis)在中科 院上海有机所君谋楼一楼报告厅成功召开,麻生明院士和唐勇研究员共同担任大 会主席。国家自然科学基金委员会化学科学部主任林国强院士,中科院上海有机 所丁奎岭所长出席了开幕式。来自上海有机所、华东师范大学、华东理工大学、 复旦大学、上海交通大学、厦门大学、中国科技大学等科研院所和高校的两百多 名专家、科研工作者以及学生参加了本次论坛。 丁奎岭所长及林国强院士先后在开幕式上致辞,表达了对与会专家、学者 的热烈欢迎和诚挚感谢。本次会议共邀请了来自美国、加拿大、德国、西班牙、 日本以及中国的 19 名著名专家作了精彩的学术报告,主要围绕金属有机试剂参 与的有机合成、金属参与或催化的有机化学反应、不对称合成与催化等前沿及交 叉科学领域进行交流与探讨,突出均相催化领域的最新发展和未来态势。整个会 议反响热烈,很多听讲的学生和科研人员都表示有这样的机会了解学科前沿的相 关知识受益匪浅,同时结合自己在科研工作过程中遇到的问题与专家进行了积极 的沟通和交流。 国际均相催化论坛是由麻生明院士发起,金属有机化学国家重点实验室主办 的每两年一次的系列国际研讨会,自 2004 年以来,已在杭州、苏州、上海、昆 明等地成功举办了四届。该系列会议旨在为来自世界各地的均相催化领域的中外 科学家搭建桥梁,提供交流化学新思想、新成果的机会,共同商讨均相催化领域 的进一步发展计划,促进学术交流,探讨及建立合作。 四、实验室学术委员会工作 金属有机化学国家重点实验室第三届学术委员会第三次会议于 2012 年 10 月 23-24 日在北京召开。国家自然科学基金委化学部常务副主任梁文平研究员、 杜灿屏研究员、郑企雨处长以及中国科学院高技术研究与发展局化工处唐清研究 员应邀参加了此次会议。 在会议上,实验室主任唐勇研究员汇报了重点实验室 2012 年度工作进展, 四川大学的冯小明教授作了题为“手性氮氧金属络合物催化不对称反应研究”的专 8 家报告,北京大学的焦宁教授作了题为“Aerobic Oxidation and Oxygenation”的学 术动态介绍,刘磊教授、王春江教授、施敏研究员、肖文精教授、唐勇研究员分 别汇报了各自的课题组工作进展及研究计划,此外,游书力研究员、段伟良副研 究员、陈耀峰研究员以及施章杰教授分别进行了有关 International Conference "Catalysis in Organic Synthesis"、12th International Symposium for Chinese Organic Chemists & 9th International Symposium for Chinese Inorganic Chemists 、 XXV International Conference on Organometallic Chemistry 以 及 Innovation on Organic Synthesis: The 1st Symposium in Direct C-H Activation 的国际会议介绍。 10 月 24 日上午,与会的领导、专家、学术委员以及老师等进行了热烈的讨 论和充分的交流。大家充分肯定了金属有机化学国家重点实验室近年来取得的成 绩,实验室在保持金属有机化学学科的优势和特色基础上,在面向国家重大需求 的研究方向如聚烯烃、氢甲酰化制丁醛等领域的新型催化剂研制方面也取得了很 好的成绩。同时,大家也对实验室的发展提出了建设性的建议和意见。希望实验
